Hasimun, Patonah Muzaki, Yoga Adi Restu Sukmawati, Ika Kurnia Zahra, Nadhira Aulia Kusriani, Herni |
Metabolic syndrome is associated with an increased risk of hypertension, diabetes, and cardiovascular disease, which are often triggered by oxidative stress and inflammation. This study evaluated the effects of Kaempferia galanga L extract (ERK) on oxidative stress and metabolic dysfunction using a fructose-induced male Wistar rat model. A total of 25 rats were divided into five groups: normal control, positive control, allopurinol group, and two ERK treatment groups at doses of 50 and 100 mg/kg body weight. All groups, except the normal control, were given 25% fructose in the drinking water and an injection of potassium oxonate (4.5 mg/kg body weight) for 28 days. Evaluation was done by measurement of glucose, triglyceride, malondialdehyde (MDA) and nitric oxide (NO) and histological analysis of aortic tissue. The results showed that ERK, especially at a dose of 100 mg/kg, significantly reduced glucose, triglyceride and MDA levels and increased NO levels. ERK administration also reduced the body weight of mice and prevented the formation of foam and inflammatory cells in aortic tissue. The total flavonoid content of ERK also supports its biological activity. This study concludes that ERK has potential as a natural therapeutic agent to treat metabolic syndrome through antioxidant and anti-inflammatory mechanisms.

Ambon banana peel (ABP) is rich in phenolic compounds yet unstable to light and heat. Foam-mat drying (FMD) protects bioactive components and facilitates the development of nutraceutical products. This study aimed to formulate and evaluate FMD granules containing ABP for their antioxidant and antihypertensive activities. Granules were prepared with ABP at 1% (F1), 2.5% (F2), and 5% (F3) w/w, combined with 6% Tween-80 and 15% maltodextrin, dried at 40 °C for 60 min, and blended with excipients. Physical characteristics, pH, flowability, and antioxidant activity (ABTS assay) were assessed. Antihypertensive effects were tested in Wistar rats induced with 2% NaCl and treated orally for 14 days; captopril (5 mg/kg BW) served as a positive control. Data were analyzed by one-way ANOVA. All formulations produced free-flowing granules with a moisture content <2% and a dissolution time <5 min. Antioxidant inhibition increased with ABP content, with F3 showing the highest activity (71.3 ± 1.4%). F3 also achieved the greatest reductions in systolic (â31%) and diastolic (â39%) blood pressure, approaching the effect of captopril. Moreover, F3 exhibits strong antioxidant activity and significant antihypertensive effects, indicating potential as a sustainable nutraceutical for the management of hypertension. Further studies on product stability and clinical efficacy are warranted.

| 1671468 | #3029 | Shallot (Allium ascalonicum): An overview of antidiabetic activity by α-glucosidase inhibition |
Ramadaini, Tiara Sumiwi, Sri Adi Febrina, Ellin |
Allium ascalonicum (shallot) is a medicinal plant with promising potential in the management of type 2 diabetes mellitus (T2DM). This review summarizes the phytochemical constituents, mechanisms of action, and existing research gaps that warrant further exploration for targeted drug development. A systematic literature search was performed using PubMed, ScienceDirect, and Google Scholar to identify relevant studies published between 2013 and 2023. Only full-text original research articles in English were included, while reviews and incomplete studies were excluded. The search strategy employed the keywords: âAllium ascalonicum OR shallot AND α-glucosidase.â The findings indicate that A. ascalonicum demonstrates significant antidiabetic activity across in vitro, in vivo, and limited human studies, primarily through inhibition of the α-glucosidase enzyme. This activity is largely attributed to quercetin and other flavonoid compounds, which play an essential role in reducing blood glucose levels and mitigating oxidative stress. Collectively, the evidence highlights the therapeutic potential of A. ascalonicum as a natural α-glucosidase inhibitor. However, further clinical investigations and comprehensive toxicity assessments are required to establish its efficacy and safety as a standardized herbal medicine and future phytopharmaceutical candidate for diabetes management.

Riasari, Hesti Rachmaniar, Revika Wahyuni, Sri |
Kencur (Kaempferia galanga L.) is a family of Zingiberaceae. Several studies have shown that kencur can help reduce inflammation because kencur is known to contain anti-inflammatory compounds, namely marker compounds from flavonoids, kaempferol. For the development of pharmaceutical preparations, research on anti-inflammatory plasters containing 96% ethanol extract, n-hexane extract, ethyl acetate extract and 70% ethanol extract from ginger rhizome with the addition of penetration enhancer (enhancer), namely propylene glycol. This anti-inflammatory plaster was tested for its activity in 5 groups of Wistar strain rat feet which had been induced 1% carrageenan (negative control); positive control (diclofenac sodium), ethanol96% extract, n-hexane extract, ethyl acetate extract and 70% ethanol extract from kencur rhizome and compared with plaster of kencur rhizome ethanol extract without enhancer. The results showed the effect of adding enhancers 30 minutes after administration. 96% ethanol extract and ethyl acetate extract had reduced inflammation by 79.99% in rat test animals compared to plaster ethanol extract of rhizome kencur without the addition of enhancers. Keywords : Kaempferia galanga. Basil (Ocimum sanctum L.) have a dominant content of citral compounds with antibacterial activity. Another plant suspected to contain antibacterial bioactive components is orange peel. Orange peel (Citrus sinensis L) contains phenolic compounds (phenolic acids, flavonones, and polymethoxylated flavones), carotenoids, and ascorbic acid. Phenolic compounds are known to have several activities such as antimicrobial, antioxidant, anticancer, anti-inflammatory, and anti- allergic. The aim of this research is to prove that the combination of orange peel and basil extracts has antibacterial effects against Staphylococcus aureus, which can be formulated as paper soap which is named O-Citri. This research method is experimental, involving the physical and chemical evaluation of the formulation, as well as antibacterial activity testing. The research results show that the combination of orange peel and basil extracts in the paper soap formulation has antibacterial effects. The highest concentration combination for antibacterial activity is F1 with a ratio of orange peel and basil extract at 5%:15%.

Megawati, Wanti Damayanti, Sophi |
Ursodeoxycholic acid is an active compound to treat cholesterol gallstones which is included in Biopharmaceutical Classification System class II. Ursodeoxycholic acid capsule formulations have been developed by the pharmaceutical industries. Due to requirement, active ingredient testing and dissolution tests need to be routinely carried out to control quality of the drug. Therefore, aim of this study was to develop a simultaneously analytical method for assay and dissolution test. Optimum condition of analysis were using HPLC-RID, C18 column (5 ”m; 3.9 x 300 mm), flow rate 1 mL/min, 50 ”L volume injection, acetonitrile:buffer phosphate pH 3.0 (55:45 v/v) as mobile phase, column, detector, and sample temperature were 40 oC. This method provides good linearity in a range of 50-130 % concentration for assay and dissolution test with correlation coefficient values of 1 and 0.9995, respectively. LOD and LOQ for assay were in value of 4.892 ”g/mL and 14.824 ”g/mL whereas for dissolution tests in a value of 6,501 ”g/mL and 19,701 ”g/mL. The average percent recovery for assay and dissolution test were in a range of 100.018±0.888% and 98.936±2.124 %. Repeatability and intermediate precision of assay were obtained by determining RSD values of 0.466% and 0.279-0.483 %, while the dissolution test were 1.137% and 0.032-0.289 %. The validation show this method are linear, selective, accurate, and precise. Keywords: Assay, dissolution test, Ursodeoxycholic acid, validation
